Microbial Host Engineering: beyond the Metabolic Pathway
نویسنده
چکیده
As the focus of microbial metabolic engineering shifts to include bulk commodities, two aspects of this research have become increasingly important. One is the requirement for high production titers in order to make the process economically viable. The second is to engineer microbes that are tolerant to these target products, many of which are toxic to cell growth. This review focuses on recent advances in the bacterial production of bulk products such as biofuels, the resources available to aid in targeted strain engineering and future directions being pursued for such efforts. A review of metabolic and enzymatic engineering strategies for designing and optimizing performance of microbial cell factories
Microbial cell factories (MCFs) are of considerable interest to convert low value renewable substrates to biofuels and high value chemicals. This review highlights the progress of computational models for the rational design of an MCF to produce a target bio-commodity. In particular, the rational design of an MCF involves: (i) product selection, (ii) de novo biosynthetic pathway identification ...

With the development of metabolic engineering, employment of a selected microbial host for accommodation of a designed biosynthetic pathway to produce a target compound has achieved tremendous success in the past several decades. Yet, increasing requirements for sophisticated microbial biosynthesis call for establishment and application of more advanced metabolic engineering methodologies. Rece...
متن کاملBiofuel production: an odyssey from metabolic engineering to fermentation scale-up
Metabolic engineering has developed microbial cell factories that can convert renewable carbon sources into biofuels. Current molecular biology tools can efficiently alter enzyme levels to redirect carbon fluxes toward biofuel production, but low product yield and titer in large bioreactors prevent the fulfillment of cheap biofuels. There are three major roadblocks preventing economical biofuel...
متن کاملGenome engineering for improved recombinant protein expression in Escherichia coli
A metabolic engineering perspective which views recombinant protein expression as a multistep pathway allows us to move beyond vector design and identify the downstream rate limiting steps in expression. In E.coli these are typically at the translational level and the supply of precursors in the form of energy, amino acids and nucleotides. Further recombinant protein production triggers a globa...
متن کاملDevelopment of a broad-host synthetic biology toolbox for ralstonia eutropha and its application to engineering hydrocarbon biofuel production
BACKGROUND The chemoautotrophic bacterium Ralstonia eutropha can utilize H2/CO2 for growth under aerobic conditions. While this microbial host has great potential to be engineered to produce desired compounds (beyond polyhydroxybutyrate) directly from CO2, little work has been done to develop genetic part libraries to enable such endeavors. RESULTS We report the development of a toolbox for t...
